The idea behind the 36 questions is to see if you are romantically compatible, but I've done it with friends for kicks and it definitely gave me a better understanding of them.

ric, Very good. There are actually many good "best" questions. But, the key to understanding someone is to read body language when they answer. 80% of all communication is non verbal. If I had to give a "best" question, it would be this. "Please tell me about your parents."
My first conversation with the woman that became my wife lasted about 8 hours. I'm not very good at distilling things down to a single question, but our marriage is still going strong after 15 years. The things I consider basics are: agreement on finances and children, as those two things can cause the most stress. It also helps to have agreement on religion and politics. However you get to those agreements is not as important as sticking with them. If you can't stick with it; find someone more compatible so that all three of you (the person you are now leaving, yourself, and the person you will find) will have a happier life.

What convinced me to marry my wife (now ex-wife) was the way she ate. Plus she was hot, but she ate like a man. I had to marry that.

I think the best way to get to know someone isn't to interrogate them with questions but to have a back and forth conversation and let the questions be part of the natural flow.
Listen to what they have to say and work with that. Offer some insights into yourself, without turning the conversation to be all about you.
